Anyone used/using Wordpress MU?

Does it mean i can run 100 minisites, all different themes, plugins, from one Wordpress MU install?
 
Pretty much yes.

I have Wordpress MU setup on my server. Currently run about 8 sites of the 1 install. Is good for saving space.

Point to note is that Wordpress MU has/is going to stop being supported by Wordpress. They seem to be focusing on Buddypress instead.

Not quite, buddypress is only a feature-rich plugin for wordpressmu.

The core features of wordpress mu will be merged into wordpress from ~wp3.0 onwards.

Depends on your server - you can use a plugin called domain map to configure multi-domains.
If you are on a shared server the http conf file will have a fit though:twisted:
